POSTDOCTORAL POSITION

Department of Microbiology
Boston University School of Medicine

A
POSTDOCTORAL POSITION is available immediately to study the role of nuclear receptors as inhibitors of HIV-1 mucosal transmission to women.

Inflammatory and ulcerative sexually transmitted infections (STIs) enhance heterosexual transmission of HIV-1. This enhanced transmission is, in part, due to recruitment of HIV-1 target cells such as Langerhans/dendritic cells (LC/DC), macrophages (MØ)‚ and T lymphocytes to sites of inflammation, and to direct activation of target cells by STIs. A common feature of STI pathogens is that they encode ligands for members of the Toll-like receptor (TLR) family of pattern recognition receptors and these ligand-activated TLRs can both activate HIV-1 target cells and induce local inflammatory responses. Importantly, ligand-activated nuclear receptors (NR), including peroxisome proliferator activated receptor (PPAR), liver X receptor (LXR), glucocorticoid receptor (GR), have been shown to be potent inhibitors of TLR-induced inflammatory gene expression. A major goal of the laboratory is to examine the potential role of ligand-activated NR as inhibitors of HIV-1 transmission. To achieve these goals, we are:

1) evaluating the impact of NR/TLR crosstalk on HIV-1 replication and inflammatory gene expression in primary LC, DC, MØ and T cells
2) determining the mechanism(s) of TLR-modulated HIV-1 transcription and how it is regulated by NR signaling
3) examining the effects of NR/TLR crosstalk on HIV-1 infection of target cells and inflammation in vaginal and cervical tissue explants and in an organotypic model of the human vagina.

Experience in cell and molecular biology and virology is desirable but not essential. The successful candidate will be part of a vibrant and highly collaborative community of immunologists and virologists, including a number of basic science and clinical HIV-1 virologists. A detailed description of the Department of Microbiology and the training programs in Immunology and Host/Pathogen Interactions can be found at [EXTERNAL]http://www.bumc.bu.edu/microbiology/ [EXTERNAL]

POSTDOCTORAL POSITION

Molecular Genetics and Pathogenesis of Bacteroides fragilis
Department of Microbiology and Immunology
Brody School of Medicine, East Carolina University

An immediate opportunity exists for postdoctoral research on the indigenous, intestinal tract anaerobe, Bacteroides fragilis, a phylogenetically novel organism that is responsible for a variety of opportunistic infections. An important virulence factor for B.fragilis infections is their remarkable aerotolerance which is controlled by an extensive oxidative stress response. Expression of nearly 40% of the genome is modulated by exposure to oxygen and this results in the development of a metabolic state that is resistant to a variety of environmental stresses. It is our goal to identify unique regulatory genes that mediate this transcriptional response and determine their role in virulence using an intra-abdominal abscess model system. The incumbent will characterize these regulatory genes and their regulons using a range of molecular genetic approaches. Current efforts are focused on the analysis of several ECF-like sigma factors involved in the transcription of a distinct subset of oxidative stress response genes.
